麻黄素对体外培养的人鼻腔纤毛运动的影响

目的观察不同浓度麻黄素对体外培养的人鼻腔纤毛运动的影响。方法采用组织块培养法,观察在加有羟乙基哌嗪乙磺酸[4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid,HEPES]的Hank平衡盐溶液(Hank balanced salt solution,HBSS)、2.5 g/L、5g/L、10g/L和20g/L的麻黄素作用下体外培养人鼻腔纤毛摆动频率(ciliary beat frequency CBF),并进行配对分析。结果①CBF在HBSS中10 min内无显著性变化;②在加入2.5 g/L、5 g/L、10 g/L和20 g/L的麻黄素后,1~2 min内CBF明显升高并达高峰,与加药前相比差异有统计学意义(P值均<0.05),后渐呈下降趋势,在10 min时2.5g/L和5g/L的麻黄素中CBF与加药前相比虽略低,但差异无统计学意义(P值均>0.05),而10 g/L和20 g//L麻黄素中CBF与加药前相比明显降低,且差异有统计学意义(P值均<0.05);③2.5 g/L~20 g//L的麻黄素作用1 min后CBF的增加量差异无统计学意义(F=0.697, P>0.05),10 min后随着浓度的增加而下降,10 g/L~20 g/L麻黄素CBF的下降值较2.5 g/L~5g/L的下降值差异有统计学意义(P<0.05)。结论麻黄素作为肾上腺素受体激动药物,2.5 g/L~20 g/L的麻黄素对体外培养的人鼻腔纤毛运动早期有一定的刺激作用,后期则表现为抑制作用,且随着药物浓度的增加,刺激作用逐渐缩短,抑制作用逐渐延长。5g/L麻黄素作用于鼻腔纤毛细胞刺激作用最大,而抑制作用较弱。 Objective To observe the effects of different concentrations of ephedrine on the cilia in human nasal cavity cultured in vitro. Methods The Hank balanced salt solution (HBSS) supplemented with [4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid, HEPES], 2 was observed by tissue culture. The ciliary beat frequency (CBF) of human nasal cilia was cultured in vitro under the action of .5 g/L, 5 g/L, 10 g/L and 20 g/L ephedrine, and paired analysis was performed. Results 1 There was no significant change in CBF within 10 min in HBSS. 2 CBF increased significantly within 1 to 2 min after adding 2.5 g/L, 5 g/L, 10 g/L, and 20 g/L ephedrine. High and high peaks, compared with pre-dosing, the difference was statistically significant (P <0.05), then gradually decreased, and CBF at 2.5 g/L and 5 g/L ephedrine at 10 min. Although it was slightly lower than that before dosing, the difference was not statistically significant (all P>0.05), but CBF in 10 g/L and 20 g/L ephedrine was significantly lower than that before dosing. The difference was statistically significant (P<0.05); there was no significant difference in the increase of CBF after 1 min of 32.5 g/L~20 g/L ephedrine (F=0.697). (P>0.05). After 10 minutes, the concentration of ephedrine decreased as the concentration increased. The decrease in the ephedrine CBF between 10 g/L and 20 g/L was lower than the decrease between 2.5 g/L and 5 g/L. Statistical significance (P<0.05). Conclusion Ephedrine is an adrenergic receptor agonist and 2.5 g/L to 20 g/L of ephedrine has a certain stimulatory effect on early human ciliated muscle movement in vitro, and it shows inhibitory effect in the later period. With the increase of drug concentration, the stimulatory effect was gradually shortened, and the inhibitory effect was gradually extended. The effect of 5g/L ephedrine on nasal cilia cells was maximal, but inhibition was weak.
