目的 :研究绿色荧光蛋白 (GFP)在前列腺癌 PC- 3M细胞中表达及对其细胞周期的影响。方法 :制备含绿色荧光蛋白基因的重组 DNA,用阳离子脂质体 Lipofectin Regant转染培养的 PC-3M细胞 ,在倒置荧光显微镜下观察 GFP的表达情况 ,同时经流式细胞仪测定阳性克隆的细胞周期。结果 :转染后有 1 0 %~ 1 5%细胞表达绿色荧光蛋白 ,且阳性克隆细胞与亲本细胞的细胞周期无明显改变。结论 :GFP是研究活细胞生命现象的一种新途径
Objective: To study the expression of green fluorescent protein (GFP) in prostate cancer PC-3M cells and its effect on cell cycle. Methods: Recombinant DNA containing green fluorescent protein (EGFP) gene was prepared. The cultured PC-3M cells were transfected with Lipofectin Regant. The expression of GFP was observed under an inverted fluorescence microscope. The positive clones were determined by flow cytometry cycle. Results: After transfection, 10% ~ 15% of the cells expressed green fluorescent protein, and the cell cycle of the positive clonal cells and the parental cells did not change significantly. Conclusion: GFP is a new way to study living cell life phenomena
