目的:研究肿瘤相关基因NGX6在胃癌与大肠癌组织以及其配对癌旁正常组织中的表达,探讨此基因在胃癌、大肠癌发生发展中的作用.方法:采用RT-PCR方法、点杂交、Northern-blot方法分别检测34例胃癌、34例大肠癌患者癌瘤组织、癌旁(手术切缘,距肿瘤5cm以上)正常组织NGX6基因表达.结果:RT-PCR显示:NGX6基因在73.5%的大肠癌组织(25/34)和93.8%的有淋巴结或远处转移的大肠癌组织(15/16)中存在表达减弱或不表达,其表达下调率明显高于相应的癌旁组织和无淋巴结或远处转移大肠癌(26.5%、55.6%,P<0.05)。但此基因表达与大肠癌病理类型无显著相关性。点杂交和Northern-blot证实了RT-PCR结果。而胃癌与配对癌旁正常组织中未发现NGX6表达水平改变(表达下调率:29.4%vs41.2%,P>0.05).结论:NGX6基因在大肠癌中表达下调,此基因的低表达在大肠癌的发生和转移中起着一定的作用,但可能与胃癌的发生无关.
OBJECTIVE: To study the expression of NGX6 in gastric cancer and colorectal cancer tissue and its adjacent normal tissues, and to explore the role of NGX6 in the development of gastric cancer and colorectal cancer.Methods: RT-PCR, dot blot, Northern blot -blot were used to detect the expression of NGX6 gene in 34 cases of gastric cancer, 34 cases of colorectal cancer and NGL6 adjacent to the tumor (surgical margin, 5 cm away from the tumor) .Results: RT-PCR showed that NGX6 was expressed in 73.5% of the large intestine The cancerous tissues (25/34) and 93.8% of colorectal cancer tissues with lymph node or distant metastasis (15/16) showed decreased expression or no expression, and the down-regulation rate was significantly higher than that in corresponding paracancerous tissues and without lymph nodes or Colorectal cancer distant metastasis (26.5%, 55.6%, P <0.05). However, this gene expression and pathological type of colorectal cancer no significant correlation. Dot blotting and Northern-blot confirmed the RT-PCR results. NGX6 expression level was not found in gastric cancer and adjacent normal tissues (down-regulation rate: 29.4% vs41.2%, P> 0.05) .Conclusion: The expression of NGX6 is down-regulated in colorectal cancer and the expression of this gene is low in large intestine Carcinogenesis and metastasis play a role, but may not be related to the occurrence of gastric cancer.
