目的分析乌鲁木齐地区献血者HPA1~17基因多态性。方法采用PCR-SSP方法对605名无血缘关系的无偿献血者进行HPA1~17系统基因分型,计算基因频率与基因型频率,并与不同人群进行比较分析。结果每份标本均表达出HPA-a基因的1a~17a基因;仅表达HPA-b基因中的1b~6b和15b;HPA7a~14a、16a、17a呈单态性;对于HPA-1、-2、-4、-5、-6主要以aa纯合子表达为多;在HPA1~17中,具有最高杂合度的是HPA-15,基因型HPA15a/15a、HPA15a/15b、HPA15b/15b频率分别是0.2760、0.5438、0.1802,其次是HPA-3,基因型HPA3a/3a、HPA3a/3b、HPA3b/3b频率分别是0.3471、0.4760、0.1769。经χ2检验,结果符合Hardy-Weinberg遗传平衡定律。结论乌鲁木齐地区HPA基因分布具有自身特点,其中HPA-15和HPA-3抗原不配合率较高。同时在此次研究数据的基础上建立了乌鲁木齐地区已知HPA基因型血小板供者数据库。
Objective To analyze the polymorphisms of HPA1-17 gene in blood donors in Urumqi. Methods PCR-SSP method was used to genotype HPA1-17 system in 605 blood donors without blood relationship, and the gene frequency and genotype frequency were calculated and compared with different populations. Results The gene 1a-17a of HPA-a gene was expressed in each specimen. Only 1b-6b and 15b of HPA-b gene were expressed. HPA7a-14a, 16a and 17a were single- , -4, -5 and -6 were mainly expressed as aa homozygotes. In HPA1-17, the highest heterozygosity was HPA-15, and the frequencies of genotypes HPA15a / 15a, HPA15a / 15b and HPA15b / 15b were 0.2760,0.5438,0.1802, followed by HPA-3. The frequencies of genotypes HPA3a / 3a, HPA3a / 3b and HPA3b / 3b were 0.3471,0.4760,0.1769 respectively. The χ2 test, the results meet the Hardy-Weinberg law of genetic balance. Conclusion The distribution of HPA gene in Urumqi has its own characteristics, of which the HPA-15 and HPA-3 antigen do not mix well. At the same time, based on the data of this study, a database of known platelet donor of HPA genotype in Urumqi was established.
