目的探讨复方中药(苦黄汤)在大鼠原位肝移植急性排斥反应模型中抑制急性排斥反应(Acute Rejection,AR)的原因。方法采用Wistar-SD原位肝移植AR模型,将40只大鼠受体分组为生理盐水组(n=20)和苦黄汤组(n=20)。从肝移植术前第1d至术后1个月,灌胃给苦黄汤(1次/d),剂量为2ml/d。术后7d处死生理盐水组大鼠和苦黄汤组大鼠各10只,评估排斥反应指数(RAI值),且分离大鼠原代肝细胞进行苦黄汤对肝移植术后AR模型中的肝细胞的研究。结果与生理盐水组对比,苦黄汤组术后生存时间显著长(P=0.016)。苦黄汤组大鼠RAI值明显低于生理盐水组。与生理盐水组相比,术后7d分离的苦黄汤组大鼠肝细胞增殖能力显著增高。结论苦黄汤可维持或增强原位肝移植AR模型大鼠肝细胞的增殖能力,且可能与抑制AR功能间存在正相关。
Objective To investigate the reason of inhibition of acute rejection (AR) in the acute rejection model of orthotopic liver transplantation in rats by compound Chinese herbal medicine (Kuhuang decoction). Methods The Wistar-SD orthotopic liver transplantation AR model was used to divide 40 rat recipients into normal saline group (n=20) and Kuhuang Decoction group (n=20). From the 1st day before liver transplantation to 1 month after surgery, Kuhuang Decoction (1 dose/d) was intragastrically administered at a dose of 2 ml/d. At 7 days after surgery, 10 rats in the normal saline group and 10 rats in the bitter yellow decoction group were sacrificed to evaluate the rejection index (RAI value). Rat primary hepatocytes were isolated and the Kuhuang Decoction was used to treat hepatocytes in an AR model after liver transplantation. Research. Results Compared with saline group, the survival time of Kuhuang Decoction group was significantly longer (P=0.016). The RAI of rats in Kuhuang Decoction group was significantly lower than that of saline group. Compared with the saline group, the proliferation of hepatocytes in the Kuhuang Decoction group was significantly increased at the 7th day after operation. Conclusion Kuhuang Decoction can maintain or enhance the proliferation ability of hepatocytes in orthotopic liver transplantation AR model rats, and it may be positively related to the inhibition of AR function.
