目的观察人乳头瘤病毒(HPV)感染子宫颈组织后蛋白质组表达情况,探讨高、低危基因型感染子宫颈组织后表达的差异及意义。方法采用比较蛋白组学研究技术,对HPV高危基因型感染的子宫颈上皮内瘤变、低危基因型感染的子宫颈尖锐湿疣和正常对照的子宫颈上皮组织进行两两比较,筛选差异表达蛋白质,并对差异蛋白质点进行质谱分析。结果对3组26个差异蛋白质点的分析共获得了22张肽质量指纹图,搜索后得到18个与其相匹配的蛋白质点。通过查询NCBInr数据库获得初步鉴定的18个蛋白质功能信息,主要与细胞代谢、信号传导、细胞分泌、细胞骨架构建及细胞增殖、分化与凋亡等相关。结论高、低危基因型HPV感染子宫颈组织后蛋白质组表达存在明显差异。
Objective To observe the proteome expression of human papillomavirus (HPV) infected cervical tissue and to explore the difference and significance of high and low risk genotype infection of cervical tissue. Methods A comparative proteomic study was carried out to compare the cervical intraepithelial neoplasia (HPV) genotype-infected cervical intraepithelial neoplasia, low-risk genotype-infected cervical condyloma acuminatum and normal control cervical epithelium tissue, and to screen the differentially expressed proteins , And differential protein spots were analyzed by mass spectrometry. Results A total of 22 peptide mass fingerprints were obtained from the analysis of 26 differential protein spots in 3 groups. After searching, 18 protein spots matched with them were obtained. The preliminary identification of 18 protein functional information obtained by querying NCBInr database was mainly related to cell metabolism, signal transduction, cell secretion, cytoskeleton construction and cell proliferation, differentiation and apoptosis. Conclusion There is a significant difference in proteomic expression between cervical cancer tissues infected with high and low risk HPV genotypes.
