以‘大五星’枇杷花药培养获得的子叶胚为材料,研究基本培养基、蔗糖浓度、4℃低温处理、饱和CaCl2脱水处理、“4℃低温+饱和CaCl2脱水”处理对胚状体成苗的影响,并优化枇杷花药培养及植株再生技术体系。结果表明:枇杷花药胚状体经“4℃低温+饱和CaCl2脱水”预处理7 d效果最佳;将预处理后的胚状体种于萌发培养基1/2MS+3%蔗糖,胚状体成苗率最高,为78.2%;将再生植株移栽入配比为腐熟有机肥∶园土∶锯末=1∶2∶1的基质中时,组培苗移栽成活率达85.25%。
The cotyledon embryos obtained from anther culture of ’Da Wu Xing’ were used as materials to study the effects of basic medium, sucrose concentration, low temperature treatment at 4 ℃, dehydration treatment with saturated CaCl2, dehydration at 4 ℃ and saturated CaCl2 Miao affect, and optimize the loquat anther culture and plant regeneration technology system.
