比较了2种低温预处理天数、诱导培养基中添加麦芽糖或蔗糖、添加较高或较低浓度生长素、愈伤组织转移时体积大小、分化培养基中添加不同浓度激素的大麦花培愈伤组织的诱导率、白苗和绿苗分化率。依据试验结果可以认为:25d左右的3~5℃大麦幼穗预处理;诱导培养基中添加3~4mg/L2,4-D、0.5~1.0mg/LNAA、80g/L麦芽糖;分化培养基中添加1.0~2.0mg/L6-BA、0.1~0.5mg/L NAA;愈伤组织体积为2~3mm时转入分化培养基等措施,有利于提高绿苗数量和减少白苗数量。
