目的对聚甲基丙烯酸缩水甘油酯(PGMA)预浸润石英纤维进行体外细胞毒性和生物相容性研究,以便对该材料应用于口腔临床提供实验依据。方法使用CCK8法测定小鼠成纤维细胞(L929)以及小鼠牙龈上皮细胞(GE1)在不同材料(非聚合状态PGMA预浸润石英纤维、聚合状态PGMA预浸润石英纤维)浸提液中培养2、4、7 d的吸光度(A值),计算细胞相对增殖率(RGR),判断细胞毒性的级别。结果非聚合状态PGMA预浸润石英纤维100%浸提液、50%浸提液细胞组在不同时间点相对增殖率为44.59%~65.66%(L929)和52.06%~62.86%(GE1),其细胞毒性程度为2~3级。而聚合状态PGMA预浸润石英纤维100%浸提液、50%浸提液细胞组的相对增殖率为90.56%~100.89%(L929)和89.99%~101.21%(GE1),其细胞毒性为0~1级。结论聚合状态PGMA预浸润石英纤维安全可靠,具有良好的临床应用前景。
OBJECTIVE To study the in vitro cytotoxicity and biocompatibility of PGMA preimpregnated quartz fibers in order to provide an experimental basis for the application of PGMA in oral clinics. Methods The CCK8 method was used to detect the growth of mouse fibroblasts (L929) and mouse gingival epithelial cells (GE1) in extracts of PGMA preimpregnated quartz fibers and PGMA preimpregnated quartz fibers in a non-polymerized state. 4,7 d absorbance (A value), calculate the relative cell proliferation rate (RGR), to determine the level of cytotoxicity. Results The relative proliferation rates of PGMA preimpregnated silica fiber 100% and 50% leachate cells at different time points were 44.59% ~ 65.66% (L929) and 52.06% ~ 62.86% (GE1) Toxicity level of 2 to 3. The relative proliferation rates of PGMA preimpregnated silica fiber 100% leachate and 50% leachate cells were 90.56% ~ 100.89% (L929) and 89.99% ~ 101.21% (GE1), respectively. The cytotoxicity was 0 ~ Level 1. Conclusions PGMA preimpregnated quartz fiber is safe and reliable, and has good clinical application prospects.
