柠条萌芽种子经低温(4~6℃)预处理72h,然后置于25~28℃温箱回干,用TTC检测幼根生活力.处理提高6天.其原因是预处理后在回干过程中提高了幼根PrO含量5倍,一方面导致细胞液浓度增加4.1%,含水率增加一倍稍多;另一方面增强了膜的稳定性,降低了细胞相对电导率(约为对照的1/2).用薄板层析极性脂再经双波色谱扫描仪测定.预处理有利于回干过程保持磷脂稳定,回干72h与0h比较,对照磷脂减少16.98%,处理仅降低3.1%.用气相色谱分析膜脂脂肪酸组成,预处理后回干48h、18:0减少5.3%,18:2增加23.6%;尔后发生相反变化,72h18:0增加5.0%,18:1增加9.3%,18:2减少15.0%.结果表明,预处理推迟了膜对回干的反应,从而提高其抗回干能力。
Germinated seeds of Caragana korshinskii were pretreated with low temperature (4-6 ℃) for 72h and then placed in a thermostat at 25-28 ℃ for dryness. The viability of young root was detected by TTC. The treatment increased by 6 days.The reason was that after pretreatment, PrO content in rootlets increased by 5 folds during the process, which led to a 4.1% increase in cell concentration and a slight increase in water content by a factor of 2. On the other hand, the stability of the membrane was enhanced and the relative conductivity of the cells was decreased 1/2) .The TLC was used to determine the polarity of lipids and then determined by dual-wavelength chromatography.The pretreatment was conducive to the dry process to maintain the phospholipid stable, dry back 72h compared with 0h, the control phospholipid decreased by 16.98%, the treatment decreased by only 3.1% The fatty acid composition of membrane lipids was analyzed by gas chromatography. After pretreated for 48h, the ratio of 18: 0 decreased by 5.3% and that of 18: 2 increased by 23.6% 18: 2 decreased by 15.0%. The results showed that pretreatment delayed the membrane’s response to dry-out and improved its anti-dry-out ability.
