目的 :观察血管紧张素Ⅱ (AngⅡ )及缬沙坦对培养的牛主动脉血管平滑肌细胞 (VSMC)增殖、迁移的作用。方法 :取新生小牛胸主动脉采用贴块法培养平滑肌细胞 ,分成对照组、AngⅡ组、缬沙坦组、缬沙坦加AngⅡ组 ,干预后分别测定3 H 胸腺嘧啶 (3 H TdR)掺入量和计数迁移的细胞。结果 :AngⅡ (1μmol/L)作用 2 4h能使VSMC的3 H TdR的掺入量、细胞迁移数较对照组增多 ;与AngⅡ组相比 ,AngⅡ加用缬沙坦 (10 0 μmol/L)使3 H TdR的掺入量与细胞迁移数明显减少 ;与对照组相比 ,单用缬沙坦使3 H TdR的掺入量亦明显减少。结论 :AngⅡ对VSMC有促增殖、迁移作用 ,能被缬沙坦拮抗 ,而且缬沙坦在没有AngⅡ作用时亦能单独发挥抗增殖作用
Objective: To observe the effects of angiotensin Ⅱ (AngⅡ) and valsartan on the proliferation and migration of cultured aortic vascular smooth muscle cells (VSMCs). Methods: Smooth muscle cells were cultured in the thoracic aorta of neonatal calves and divided into control group, AngⅡgroup, valsartan group, and valsartan plus AngⅡgroup. The levels of 3H-thymidine Enter and count migrated cells. RESULTS: Compared with Ang Ⅱ group, Ang Ⅱ plus Valsartan (100 μmol / L) increased the amount of 3 H TdR incorporation and the number of cell migration in VSMCs after 24 h treatment with AngⅡ (1 μmol / L) The incorporation of 3 H TdR and the number of cell migration were significantly reduced. Compared with the control group, the incorporation of 3 H TdR by valsartan alone was also significantly reduced. CONCLUSION: AngⅡ can promote proliferation and migration of VSMC, and can be antagonized by valsartan. Moreover, valsartan can exert anti-proliferative effect alone without AngⅡ
