目的1-甲氧基钩吻碱对福尔马林足底注射所致炎性痛及小鼠坐骨神经慢性压榨性损伤(CCI)所致神经病理性疼痛的作用。方法建立小鼠福尔马林炎性疼痛模型,以注射福尔马林后1 h内小鼠舔足时间为指标,观察1-甲氧基钩吻碱对炎性痛第二相痛反应的影响;建立小鼠CCI模型,于术后第3天至术后第9天sc 1-甲氧基钩吻碱,每天2次,连续7 d,以双足热痛缩足潜伏期比值为指标,观察1-甲氧基钩吻碱对小鼠足底热痛缩足潜伏期的影响。结果1-甲氧基钩吻碱具有显著对抗小鼠福尔马林致炎性疼痛第二相痛反应作用,作用呈剂量依赖性,ED50为4.59 mg/kg。1-甲氧基钩吻碱具有显著的抗CCI小鼠热辐射痛敏的作用,镇痛效应呈剂量依赖性;高剂量组小鼠给药后2 d显效,镇痛疗效持续至停药后36 h;术后第9天,1-甲氧基钩吻碱镇痛效应ED50为3.84 mg/kg。结论1-甲氧基钩吻碱具有抗炎性疼痛作用与抗神经病理性疼痛作用。
OBJECTIVE: To investigate the effects of 1-methoxyglutarylin on inflammatory pain induced by formalin in plantar and chronic neuropathic pain induced by chronic sciatic nerve injury (CCI) in mice. Methods The formalin-induced inflammatory pain model was established in mice. The time of licking test in mice within 1 h after injection of formalin was used as an index to observe the effect of 1-methoxyglutarylin on the second phase pain reaction of inflammatory pain. The model of CCI was established in mice. Sc 1-methoxykiseline was administered from the third day to the ninth postoperative day, twice a day for 7 consecutive days. Effect of Methycobalosine on Thermal Shock and Fecal Induce of Mouse. Results 1-Methoxycarnitine had a significant anti-second phase pain response to formalin-induced inflammatory pain in mice with a dose-dependent ED50 of 4.59 mg / kg. 1-Methoxycarnitine had a significant anti-hyperalgesia effect on CCI mice, and the analgesic effect was dose-dependent. In high-dose group, the mice were markedly effective 2 days after dosing and the analgesic effect continued until after drug withdrawal 36 h. On the 9th day after operation, the ED50 of 1-methoxybenzamide was 3.84 mg / kg. Conclusion 1-Methoxycarnitine has anti-inflammatory pain and anti-neuropathic pain.
