目的在体外研究α2整合素受体与骨肉瘤细胞胞内基质金属蛋白酶2(MMP-2)的关系。方法用含不同浓度α2整合素单克隆抗体的DMEM培养基,培养MG63细胞6 h后,Trizol法提取细胞总RNA,RT-PCR半定量检测MMP-2的mRNA表达。分别用含浓度为10、25、50μg/ml的α2整合素单克隆抗体的DMEM培养基,培养MG63细胞48 h后,免疫细胞化学法检测MMP-2蛋白表达。结果随α2整合素单克隆抗体抗体浓度升高,MG63细胞MMP-2的mRNA和蛋白表达均降低。结论α2整合素受体可促进骨肉瘤MG63细胞MMP-2的表达,这种促进作用体现在RNA和蛋白水平。
Objective To investigate the relationship between α2 integrin receptor and intracellular matrix metalloproteinase 2 (MMP-2) in osteosarcoma cells in vitro. Methods DMEM medium containing different concentrations of α2 integrin monoclonal antibody was used to culture MG63 cells for 6 h. Total RNA was extracted by Trizol method and semi-quantitative RT-PCR was used to detect the mRNA expression of MMP-2. The MG63 cells were cultured for 48 h in DMEM medium containing α2 integrin monoclonal antibody at a concentration of 10, 25 and 50 μg / ml, respectively. Immunocytochemistry was used to detect the protein expression of MMP-2. Results With the increase of antibody concentration of integrin α2, the mRNA and protein expression of MMP-2 in MG63 cells decreased. Conclusion α2 integrin receptor can promote the expression of MMP-2 in osteosarcoma MG63 cells. The promoting effect is reflected in RNA and protein levels.
