目的探讨血小板源性生长因子C(PDGF-C)及其变体血小板源性生长因子Cb(PDGF-Cb)在肾透明细胞癌与相应的正常组织中的表达调节及其意义。方法采用RT-PCR方法检测12例原发性肾透明细胞癌及相对应的正常肾组织中的PDGF-C,并探讨PDGF-C和PDGF-C变体表达的关系。观察顺铂对肾癌细胞株PDGF-C和PDGF-CbmRNA表达的调节作用。结果与其对应的正常组织相比较,12例肾癌组织中,9例PDGF-Cb mRNA表达水平降低,10例全长PDGF-C mRNA表达明显上调,PDGF-C/-Cb的比值有11例明显上升。在mRNA水平,顺铂对PDGF-C及PDGF-Cb呈下调作用,对PDGF-Cb的下调作用呈剂量依赖性。PDGF-C的这两种剪切变体在肾癌组织及与其相应的正常肾组织中表达差异有统计学意义(P<0.05),且全长PDGF-C的表达显著升高。结论PDGF-Cb反向调节PDGF-C的分泌,对PDGF-C表达调节作用可能与肾细胞癌的致癌性相关。顺铂对肾癌细胞株PDGF-C和PDGF-Cb mRNA表达的调节作用为肾癌的致癌机理研究提供了可能的途径。
Objective To investigate the expression regulation of platelet-derived growth factor C (PDGF-C) and its variant platelet-derived growth factor Cb (PDGF-Cb) in clear cell renal cell carcinoma and its corresponding normal tissues. Methods The expressions of PDGF-C and PDGF-C in 12 cases of primary renal clear cell carcinoma and corresponding normal renal tissues were detected by RT-PCR. To observe the regulatory effect of cisplatin on the expression of PDGF-C and PDGF-Cb mRNA in renal cell carcinoma cell line. Results Compared with the corresponding normal tissues, the expression of PDGF-Cb in 9 cases decreased in 12 cases of renal cell carcinoma, and the expression of PDGF-C in 10 cases was significantly increased. The ratio of PDGF-C / Cb was significantly higher in 11 cases rise. At the mRNA level, cisplatin on PDGF-C and PDGF-Cb was down-regulated, on the down-regulation of PDGF-Cb in a dose-dependent manner. The expressions of PDGF-C and PDGF-C in both renal cell carcinoma and corresponding normal renal tissues were significantly different (P <0.05), and the expression of full-length PDGF-C was significantly increased. Conclusion PDGF-Cb may reverse the regulation of PDGF-C secretion and regulate the expression of PDGF-C, which may be related to the carcinogenesis of renal cell carcinoma. The regulatory effect of cisplatin on the expression of PDGF-C and PDGF-Cb mRNA in renal cell carcinoma cells provides a possible approach for the study of carcinogenesis of renal cell carcinoma.
