目的筛选可供体外和动物体内肿瘤抑制试验的候选癌细胞株。方法通过间接免疫荧光法,用兔抗人绒毛膜促性腺激素(hCG)多克隆免疫球蛋白(IgG)和小鼠抗hCG单克隆免疫球蛋白(IgG),对人结(直)肠癌细胞(HCT15、HCT116、KM12、WiDr、LoVo、LS-174)、前列腺癌细胞(PC3)、小细胞肺癌细胞(A549)的细胞表达hCG情况进行检测。结果被检的8个癌细胞株均对兔抗hCG多克隆免疫球蛋白(IgG)呈阳性反应。其中4株结(直)肠癌细胞(HCT115、HCT116、Lo-Vo、LS-174)和小细胞肺癌(A549)能被小鼠抗hCG单克隆免疫球蛋白染色。结论这5株能与小鼠抗hCG单克隆免疫球蛋白结合的细胞株都适用于以后的肿瘤杀伤试验。
Objective To screen candidate cancer cell lines for in vitro and in vivo tumor suppression experiments. METHODS: Indirect immunofluorescence with rabbit anti-human chorionic gonadotropin (hCG) polyclonal immunoglobulin (IgG) and mouse anti-hCG monoclonal immunoglobulin (IgG) against human colorectal cancer cells The cells (HCT15, HCT116, KM12, WiDr, LoVo, LS-174), prostate cancer cells (PC3), and small cell lung cancer cells (A549) were tested for the expression of hCG. Results The eight cancer cell lines tested positive for rabbit anti-hCG polyclonal immunoglobulin (IgG). Four of the colorectal cancer cells (HCT115, HCT116, Lo-Vo, LS-174) and small cell lung cancer (A549) were stained with mouse anti-hCG monoclonal immunoglobulin. Conclusion The five cell lines that can bind to mouse anti-hCG monoclonal immunoglobulin are suitable for the subsequent tumor killing test.
