目的:研究iNOS基因在我国食管鳞癌中的表达特点及iNOS基因表达与PCNA表达的关系,探讨iNOS基因在食管癌变中的可能作用.方法:应用原位杂交、流式细胞术对81例食管鳞癌中iNOS及PCNA基因的表达进行检测,并应用图像分析技术对原位杂交结果进行定量分析.结果:iNOSmRNA在正常食管鳞状上皮几乎无表达,在异型增生上皮近基底细胞有少量表达,而在鳞癌细胞中有明显阳性表达,癌细胞胞质呈棕黄色颗粒状.原位杂交定量分析及流式细胞术结果都显示:iNOS在正常食管上皮表达微量,在异型增生上皮明显增高,而在不同分化鳞癌中表达都较高,在低分化鳞癌中表达最高,组间差异非常显著(F=14.001,P<0.01).PCNA表达也是随分化程度的降低而明显增高,组间差异显著(F=20.393,P<0.01).且iNOS与PCNA表达呈显著正相关(r=0.292,P=0.009).结论:在食管鳞癌中iNOS基因表达显著增高,iNOS基因表达异常与食管上皮癌变密切相关.
Objective: To study the expression of iNOS gene in esophageal squamous cell carcinoma and the relationship between iNOS gene expression and PCNA expression in esophageal squamous cell carcinoma and to explore the possible role of iNOS gene in esophageal carcinogenesis.Methods: In situ hybridization and flow cytometry Squamous cell carcinoma iNOS and PCNA gene expression were detected, and image analysis techniques were used to quantitatively analyze the results of in situ hybridization.Results: iNOS mRNA was almost no expression in normal esophageal squamous epithelium, a small amount of expression in the basal cells of dysplastic epithelium, While in squamous cell carcinoma cells were significantly positive expression, the cytoplasm of cancer cells was brown granular.Quantitative in situ hybridization analysis and flow cytometry results show that: iNOS expression in normal esophageal epithelium trace in dysplasia epithelium was significantly higher, But higher in differentiated squamous cell carcinoma and highest expression in poorly differentiated squamous cell carcinoma (F = 14.001, P <0.01) .PCNA expression was also significantly increased with the differentiation degree decreased, (R = 0.292, P = 0.009) .Conclusion: The expression of iNOS gene is significantly increased in esophageal squamous cell carcinoma, and the expression of iNOS gene is abnormally correlated with esophagus Skin cancer are closely related.
