以番茄活体植株为试材,采用高效液相色谱(HPLC)法研究了番茄活体再生中反式玉米素含量的变化。结果表明:在愈伤组织的形成过程中,反式玉米素逐渐增加,并且在愈伤组织最发达时达到峰值。随着愈伤组织分化,其含量略有降低。此变化规律与番茄离体再生中外源反式玉米素使用浓度由高到低的变化规律基本吻合。活体再生中,用细胞分裂素合成抑制剂洛伐他丁处理伤口不影响再生进程。
The living tomato plants were used as materials to study the changes of trans-zeatin content in vivo regeneration by high performance liquid chromatography (HPLC). The results showed that during the formation of callus, trans-zeatin increased gradually and reached its peak when the callus was the most developed. With callus differentiation, its content slightly reduced. The variation of this pattern was in good agreement with the change of high and low concentration of exogenous trans-zeotin in tomato in vitro regeneration. In vivo regeneration, lovastatin treatment with a cytokinin inhibitor does not affect the regeneration process.
