目的:观察氧化苦参碱(oxymatrine,OM)与氟尿嘧啶(fluorouracil,5-FU)联合应用对结肠癌LoVo细胞的影响及其作用机制。方法:MTT法测定单用OM或5-FU及两者联用对LoVo细胞的毒性作用的影响,并计算合并指数;流式细胞仪检测OM与5-FU联用对LoVo细胞凋亡的影响。结果:5-FU联用IC_(10)浓度的OM时对LoVo细胞的抑制作用比单用5-FU时增效2.67倍,OM联用IC10浓度的5-FU时比单用OM时增效2.01倍,两药的合并指数为0.87;5-FU和OM联用后,细胞凋亡率比单用时分别增加了2.08及1.82倍。结论:氧化苦参碱与氟尿嘧啶体外联用有协同抗结肠癌LoVo细胞的作用。
OBJECTIVE: To observe the effect of oxymatrine (OM) combined with fluorouracil (5-FU) on colon cancer LoVo cells and its mechanism. Methods: MTT assay was used to determine the effect of OM alone or 5-FU on the cytotoxicity of LoVo cells and the combination index was calculated. Flow cytometry was used to detect the effect of OM combined with 5-FU on the apoptosis of LoVo cells . Results: The inhibitory effect of 5-FU with IC_ (10) OM on LoVo cells was 2.67 folds higher than that of 5-FU alone, and the effect of OM with IC10 at 5-FU was greater than that of OM alone 2.01-fold, the combination of the two drugs index was 0.87; 5-FU and OM combination, the apoptosis rate increased by 2.08 and 1.82 times, respectively, when used alone. Conclusion: Oxymatrine combined with fluorouracil has synergistic effect against colon cancer LoVo cells in vitro.
