目的观察131I-GMCSF在诱导HL60/ADM细胞凋亡过程中细胞周期的改变,进一步探讨凋亡与细胞周期改变的关系。方法采用体外放射免疫治疗模型,通过流式细胞术观察131I-GMCSF诱导HL60/ADM细胞凋亡情况及其对HL60/ADM细胞周期的改变。结果Annexin V/PI双染色法证实一定放射性浓度的131I-GMCSF能明显诱导HL60/ADM细胞凋亡,且随着作用时间的延长,细胞凋亡率增加,流式细胞术显示作用24h后的细胞周期改变主要表现为S期阻滞和G2/M期阻滞,而作用48h后则主要引起细胞发生G2/M期阻滞。结论在放射免疫导向治疗中,辐射诱导的细胞凋亡与细胞周期发生阻滞,特别是与G2/M期阻滞有着密切的关系,细胞周期阻滞与放射敏感性可能存在正相关。
Objective To observe the changes of cell cycle of 131I-GMCSF in inducing HL60 / ADM cell apoptosis and to further explore the relationship between apoptosis and cell cycle changes. Methods The in vitro radioimmunotherapy model was used to observe the apoptosis of HL60 / ADM cells induced by 131I-GMCSF and its effect on the cell cycle of HL60 / ADM cells by flow cytometry. Results Annexin V / PI double staining confirmed that 131I-GMCSF with certain radioactive concentration could obviously induce the apoptosis of HL60 / ADM cells. With the prolongation of the action time, the apoptosis rate was increased. Flow cytometry showed that the cells treated with 131I- Periodic changes mainly manifested as S phase arrest and G2 / M phase arrest, while the role of 48h after the main cause of cell G2 / M phase arrest. Conclusion In radioimmunotherapy, radiation-induced apoptosis and cell cycle arrest, especially with the G2 / M phase block has a close relationship, cell cycle arrest and radiosensitivity may have a positive correlation.
