目的:探讨表皮生长因子(epiderm algrowth factor,EGF)与胰岛素对小鼠体外受精(in vitro fertilization,IVF)胚胎植入前发育的影响。方法:小鼠IVF胚胎培养在添加不同浓度的EGF和/或胰岛素-葡萄糖的CZB培养基中,于hCG注射后138h检测囊胚发育率及每个囊胚的细胞数目。结果:培养基中添加0.1ng/mlEGF显著提高了小鼠扩张囊胚的形成率,而囊胚的细胞数目并未增加。EGF与胰岛素-葡萄糖共同添加对小鼠IVF囊胚发育率及囊胚细胞数目并未有显著提高。结论:EGF通过刺激细胞分化,而非细胞增殖方式促进小鼠IVF胚胎的早期发育。而各自最佳浓度的EGF与胰岛素共同添加对小鼠早胚并未有协同促进作用。
Objective: To investigate the effects of epidermal growth factor (EGF) and insulin on the development of mouse embryos before in vitro fertilization (IVF) implantation. METHODS: Mouse IVF embryos were cultured in CZB medium supplemented with different concentrations of EGF and / or insulin-glucose. The blastocyst rate and number of blastocysts per cell were detected at 138 h after hCG injection. Results: The addition of 0.1ng / ml EGF in the medium significantly increased the rate of expansion of blastocysts in mice, while the number of blastocysts did not increase. The combination of EGF and insulin-glucose did not significantly improve the rate of blastocyst development and the number of blastocysts in mice. Conclusion: EGF promotes the early development of mouse IVF embryos by stimulating cell differentiation rather than cell proliferation. The best concentration of EGF and insulin co-added on the early embryo of mice did not synergistically promote the role.
