该文利用生物信息学的方法和工具,对5种北方药用植物萜类合酶中异戊烯焦磷酸异构酶(isopentenyl pyrophosphate isomerase,IPI)的理化性质、亲/疏水性、跨膜结构域、二级结构、亚细胞定位等方面进行分析及预测。结果表明:5种北方药用植物IPI蛋白的分子量、等电点、不稳定指数、脂肪指数等理化性质无太大差异;主要表现为亲水性;亚细胞定位大多定位于叶绿体上;磷酸化位点最多的是丝氨酸;二级结构以α-螺旋和不规则卷曲为主;均无信号肽,由此推测IPI是非分泌性蛋白;无跨膜结构域存在;主要有1个功能结构域(Nudix Hydrolase Superfamily)。该研究结果为进一步研究IPI基因的功能以及对于北方药用植物的深入研究、提高药用植物药效以及合理开发利用药用植物资源具有重要意义。
In this paper, the physicochemical properties, affinity / hydrophobicity, transmembrane structure of isopentenyl pyrophosphate isomerase (IPI) in terpenoid synthases of five northern medicinal plants were studied using bioinformatics methods and tools. Domain, secondary structure, subcellular localization and other aspects of analysis and prediction. The results showed that there were no significant differences in physicochemical properties such as molecular weight, isoelectric point, instability index and fat index of IPI proteins of the five northern medicinal plants, mainly showing hydrophilicity, subcellular localization mostly located in chloroplast, phosphorylation Serine was the most common site; the secondary structure was dominated by α-helix and irregular curl; no signal peptide was found, suggesting that IPI was a non-secreted protein; no transmembrane domain existed; there were 1 functional domain Nudix Hydrolase Superfamily). The results of this study are of great significance for the further study of the function of IPI genes and in-depth study of medicinal plants in northern China, improving the efficacy of medicinal plants and rational exploitation and utilization of medicinal plant resources.
