目的 探讨20 个植物药对肿瘤细胞DNA多聚酶的作用及对肿瘤细胞生长的影响。方法 自对数生长期的小鼠艾氏腹水癌细胞中提取核酶液,根据Ono 法,以3H-dTTP掺入活化小牛胸腺DNA的量作为DNA 多聚酶活性指标。用MTT法测定药物对肿瘤细胞生长的影响。结果 巴马汀和小蘖碱抑制小鼠艾氏腹水癌细胞DNA 多聚酶,在100 μg/m l浓度下,巴马汀抑酶率为68.5 % ±6.0 % ,小蘖碱为68.6 % ±15.3 % ,其余18 个植物药均不抑制DNA多聚酶。小蘖碱对体外培养的人早幼粒细胞白血病HL60 细胞和人类淋巴母细胞CCRF-CEM 细胞均有抑制作用,IC50 分别为5.14 μg/m l和34.55 μg/m l。结论 巴马汀和小蘖碱均是肿瘤细胞DNA多聚酶的抑制剂。小蘖碱对体外培养的肿瘤细胞有抑制作用。对DNA 多聚酶的抑制作用可能是其抗瘤机制之一。以DNA多聚酶为靶点筛选抗癌药物可作为现有筛选方法的补充
Objective To investigate the effects of 20 plant drugs on DNA polymerase in tumor cells and their effects on tumor cell growth. Methods The ribozyme solution was extracted from Ehrlich ascites carcinoma cells in the logarithmic growth phase. According to the Ono method, the amount of activated calf thymus DNA spiked with 3H-dTTP was used as an indicator of DNA polymerase activity. MTT assay was used to determine the effect of drugs on tumor cell growth. Results Palmatine and berberine inhibited DNA polymerase activity in Ehrlich ascites carcinoma cells. At a concentration of 100 μg/ml, the anti-enzyme activity of palmatine was 68.5% ± 6.0%, and that of berberine was 68.6 % ± 15.3%. The remaining 18 botanical drugs did not inhibit DNA polymerase. Berberine had inhibitory effects on human promyelocytic leukemia HL60 cells and human lymphoblast CCRF-CEM cells in vitro with IC50 of 5.14 μg/ml and 34.55 μg/ml, respectively. Conclusion Both palmatine and berberine are inhibitors of DNA polymerase in tumor cells. Berberine inhibits in vitro cultured tumor cells. Inhibition of DNA polymerase may be one of its anti-tumor mechanisms. Screening anticancer drugs using DNA polymerase as a target can complement existing screening methods
