目的初步探究甘草酸二铵(DG)对丙型肝炎病毒相关性淋巴瘤(HCV-NHL)患者单核细胞PD-1表达的免疫调控作用。方法从健康人、单纯丙型肝炎病毒(HCV)感染者和HCV-NHL患者的外周血中分离外周血单个核细胞(PB-MC),体外培养刺激后,应用流式细胞仪分析健康人、单纯HCV感染者和HCV-NHL患者CD14+PD-1+细胞占CD14+总单核细胞的比例;HCV-NHL患者PBMC加入DG和阳性对照HCV核心抗原干预后较HCV-NHL空白对照CD14+PD-1+细胞占CD14+总单核细胞比例的变化情况。结果与健康人比较,单纯HCV感染者和HCV-NHL患者CD14+PD-1+/CD14+均明显升高,并且HCV-NHL患者较单纯HCV感染者升高明显,各组之间差异均有统计学意义(P<0.05)。HCV-NHL+HCV核心抗原组CD14+PD-1+/CD14+比例较HCV-NHL空白对照明显增加,而HCV-NHL+DG组CD14+PD-1+/CD14+比例较HCV-NHL+HCV核心抗原组明显降低,各组之间差异均有统计学意义(P<0.05)。结论 HCV-NHL患者单核细胞程序性死亡通路激活增加,且程度重于单纯HCV感染,DG可以降低HCV-NHL患者单核细胞PD-1+的高表达。
Objective To investigate the immunomodulatory effect of diammonium glycyrrhizinate (DG) on PD-1 expression in monocytes of patients with hepatitis C virus-associated lymphoma (HCV-NHL). Methods Peripheral blood mononuclear cells (PBMCs) were isolated from the peripheral blood of healthy individuals, patients with HCV and HCV-NHL. After stimulated by PBMC, The ratio of CD14 + PD-1 + cells to total CD14 + monocytes in HCV-HCV patients and HCV-NHL patients was significantly higher than that in HCV-NHL blank control cells treated with HCV core antigen. 1 + cells accounted for the proportion of CD14 + total mononuclear cells. Results Compared with healthy controls, both CD14 + PD-1 + / CD14 + in patients with HCV infection and HCV-NHL were significantly higher than those in HCV-NHL patients and HCV-NHL patients with HCV infection were significantly higher than those in HCV-NHL patients Significance (P <0.05). The ratio of CD14 + PD-1 + / CD14 + in HCV-NHL + HCV core antigen group was significantly higher than that in HCV-NHL blank control group, but the ratio of CD14 + PD-1 + / CD14 + Group was significantly lower, the differences between the groups were statistically significant (P <0.05). Conclusion The activation of monocyte programmed cell death pathway in HCV-NHL patients is more severe than that of HCV infection alone. DG can reduce the expression of PD-1 + in mononuclear cells of HCV-NHL patients.
