目的观察系统性红斑狼疮(SLE)患者外周血中α干扰素(IFN-α)、白介素10(IL-10)、转化生长因子β(TGF-β)的表达变化,并探讨其意义。方法采用双抗体夹心ELISA法检测98例SLE患者(SLE组)及55例健康人(对照组)血清中的IFN-α、IL-10、TGF-β,采用RT-PCR法检测外周血单个核细胞(PBMCs)中的IFN-α、IL-10、TGF-βmRNA。结果SLE组血清中的IFN-α、IL-10及PBMCs中的IFN-α、IL-10 mRNA均高于对照组(P<0.05),且活动期高于稳定期(P<0.05);SLE组血清中的TGF-β及PBMCs中的TGF-βmRNA低于对照组(P<0.05),且活动期低于稳定期(P<0.05);SLE组血清及PBMCs中的IFN-α、IL-10及其mRNA呈正相关(r=0.80、0.75,P均<0.05),IL-10、TGF-β及其mRNA呈负相关(r=-0.45、-0.25,P均<0.05),IFN-α、TGF-β及其mRNA无相关性。SLE组血清及PBMCs中IFN-α、IL-10、TGF-β与其相应的mRNA呈正相关(r=0.90、0.82、0.91,P均<0.05)。结论SLE患者外周血中IFN-α、IL-10高表达,TGF-β低表达;三者相互作用参与SLE的发病。
Objective To observe the changes of the expression of IFN-α, IL-10 and TGF-β in peripheral blood of patients with systemic lupus erythematosus (SLE) and to explore its significance. Methods Serum levels of IFN-α, IL-10 and TGF-β in 98 patients with SLE (SLE group) and 55 healthy people (control group) were detected by double antibody sandwich ELISA. The levels of peripheral blood mononuclear cells IFN-α, IL-10, TGF-β mRNA in cells (PBMCs). Results The levels of IFN-α and IL-10 in serum of IFN-α, IL-10 and PBMCs in SLE group were significantly higher than those in control group (P <0.05) The levels of TGF-βmRNA in TGF-β and PBMCs were lower than those in control group (P <0.05), and the activities of TGF-βmRNA in serum and PBMCs were lower than those in control group (P <0.05) 10 and its mRNA (r = 0.80,0.75, P <0.05), IL-10, TGF-β and its mRNA were negatively correlated (r = -0.45, -0.25, P all <0.05) , TGF-β and its mRNA no correlation. The levels of IFN-α, IL-10 and TGF-β in serum and PBMCs of SLE group were positively correlated with their corresponding mRNA (r = 0.90,0.82,0.91, P <0.05). Conclusions The levels of IFN-α, IL-10 and TGF-β in peripheral blood of patients with SLE are high, and their interaction is involved in the pathogenesis of SLE.
