目的 观察不同发育阶段大鼠海马的形态学变化及海马CA2~ 3区nNOS的表达。方法 取大鼠2 8只 ,按鼠龄大小分为 1周龄、3周龄、3月龄及老龄 (2 4~ 2 8月龄 ) 4个年龄组 ,分别观察不同发育阶段大鼠海马形态学特征和免疫组化ABC法对海马CA2~ 3区nNOS的表达定位及定量分析。结果 不同发育阶段大鼠海马结构呈现明显的形态学变化 ;nNOS在各年龄组大鼠海马CA2~ 3区中均有表达 ,阳性细胞积分光度值 (IOD)以 3周龄组为最高 ,其次为 3月龄组 ,而 1周龄和老龄组大鼠最低。结论 不同发育阶段大鼠海马CA2~ 3区nNOS的表达表现出明显的动态变化特征 ;提示一氧化氮 (NO)与海马的发育、突触的形成和成熟以及学习记忆具有密切关系
Objective To observe the morphological changes of hippocampus and the expression of nNOS in hippocampal CA2 ~ 3 in different developmental stages. Methods Twenty-eight male SD rats were divided into four groups (1 week, 3 weeks, 3 months and 24-48 months old) according to their age. The hippocampal formation of rats at different developmental stages Immunohistochemical ABC Method and Immunocytochemical Localization of nNOS in CA2 ~ 3 Region of Hippocampus and Its Quantitative Analysis. Results The hippocampal formation of rats in different developmental stages showed obvious morphological changes. The expression of nNOS in CA2 ~ 3 of hippocampus of rats of all age groups was positive. The IOD of positive cells was the highest in 3 weeks group, followed by 3 month old group, while 1 week old and old rats were the lowest. Conclusions The expression of nNOS in hippocampal CA2 ~ 3 of rats at different developmental stages showed obvious dynamic changes. It suggests that nitric oxide (NO) is closely related to the development of hippocampus, the formation and maturation of synapse and learning and memory
