河豚毒素主要存在于河豚鱼中,是一种致命的神经毒素。将量子点标记于河豚毒素,并将其作为示踪物,获得高且稳定的荧光信号,建立荧光偏振免疫分析技术检测河豚毒素。所建立的方法检测河豚毒素检测范围为1.36~101.27μg/L,IC50值为11.76μg/L,检测限为0.67μg/L。在该方法的检测范围内标准平均回收率为(99.91±2.60)%,变异系数为2.60%。与竞争性ELISA相比较,本方法河豚毒素测定值与竞争性ELISA测定值呈线性相关,且具有简单、快速等特点,可应用于临床诊断和食品安全检测。
Tetrodotoxin is mainly found in puffer fish and is a deadly neurotoxin. Quantum dots were labeled with tetrodotoxin, which were used as tracer to obtain high and stable fluorescence signal. The fluorescence polarization immunoassay was used to detect tetrodotoxin. The established method for detection of tetrodotoxin ranged from 1.36 to 101.27 μg / L with IC50 of 11.76 μg / L with a detection limit of 0.67 μg / L. The standard average recovery within this test range was (99.91 ± 2.60)% with a coefficient of variation of 2.60%. Compared with the competitive ELISA, the measured value of tetrodotoxin in this method is linearly correlated with the competitive ELISA, and has the characteristics of simplicity and rapidness. It can be used in clinical diagnosis and food safety testing.
