目的研究桔梗种子的最适发芽条件及储藏条件。方法利用不同光照时长、不同发芽温度、不同试剂的浓度梯度、不同发芽床、不同储存温度等处理,对桔梗种子进行发芽试验,测定其发芽率、发芽势、发芽指数。结果在25℃、12h光照条件下桔梗种子发芽率最高,为90.83%;赤霉素能显著促进桔梗种子萌发,200mg/L为最佳浸种浓度,高浓度的高锰酸钾、聚乙二醇均不同程度的抑制桔梗种子的萌发,细胞分裂素起显著的抑制作用;不同发芽床对桔梗种子萌发影响显著;-20℃储存桔梗种子的发芽率最高,为94.44%。结论桔梗种子的最佳发芽条件为25℃、12h光照条件下用滤纸培养;最佳试剂为200mg/L的赤霉素;最适储存温度为-20℃。
Objective To study the optimum germination conditions and storage conditions of Platycodon grandiflorum seeds. Methods Germination tests of Campanulaceae seeds were carried out with different light durations, different germination temperatures, concentration gradients of different reagents, different germination beds and different storage temperatures. The germination rate, germination potential and germination index of the seeds were determined. Results The germination rate of Campanulaceae seeds was the highest at 90 ℃ for 12 hours under the conditions of 25 ℃ and light illumination of 12 hours. Gibberellin could significantly promote the germination of Campanulaceae seeds. The optimum concentration was 200 mg / L, potassium permanganate and polyethylene glycol All inhibited the germination of Platycodon grandiflorum seeds to a certain extent and the cytokinin played a significant inhibitory effect. The germination rate of Platycodon grandiflorum was significantly different with different germination beds, and the germination rate was 94.44% at -20 ℃. Conclusion The best germination condition of Campanulaceae seeds is 25 ℃, and it is cultured with filter paper under 12h illumination. The optimal reagent is 200mg / L gibberellin. The optimum storage temperature is -20 ℃.
