目的: 探讨哮喘患者PBMCs中转录因子T bet/GATA 3的比率与Th1 /Th2细胞失衡的关系。方法: 采用RT PCR法测定 30例发作期哮喘患者 (哮喘组 )及 20例慢性阻塞性肺病患者 (COPD组 )及 20例正常人 (对照组 )PBMCs中T bet和GATA 3的活性, ELISA法测定血浆IL 4、IL 5、IL 13和IFN γ的表达水平。结果: 哮喘组PBMCs中T bet/GATA 3的比率较COPD组和对照组明显降低, Th2细胞因子IL 4、IL 5、IL 13的水平增高, 与T bet/GATA 3的比率成负相关, 而Th1细胞因子IFN γ的水平降低, 与T bet/GATA 3的比率成正相关。结论: 哮喘患者T bet/GATA 3的比率降低, 可以作为评价Th1 /Th2细胞失衡的精确指标。
Objective: To investigate the relationship between the ratio of T bet / GATA 3 and Th1 / Th2 cell imbalance in PBMCs of asthmatic patients. Methods: The T bet and GATA 3 activities in PBMCs from 30 patients with asthma (asthma group), 20 patients with chronic obstructive pulmonary disease (COPD group) and 20 healthy controls (control group) were determined by RT-PCR. Plasma IL 4, IL 5, IL 13 and IFN γ expression levels were measured. Results: The ratio of T bet / GATA 3 in asthmatic PBMCs was significantly lower than that in COPD and control groups, and the levels of Th2 cytokines IL 4, IL 5 and IL 13 were increased, but negatively correlated with the ratio of T bet / GATA 3 Th1 cytokine IFN-γ levels decreased, and T bet / GATA 3 ratio was positively correlated. Conclusions: The decreased ratio of T bet / GATA 3 in asthma patients can be used as an accurate indicator to evaluate the imbalance of Th1 / Th2 cells.
