目的 :探讨苯并 (a)芘在人肺癌发生中的作用。方法 :用苯并 (a)芘的代谢产物反式 -BPDE ,作用于体外培养的人胎气管上皮细胞 8~ 1 2周后 ,用PCR -SSCP方法检测 p53抑癌基因第 6,7,8外显子的点突变情况。 结果 :经反式 -BPDE处理的人胎气管上皮细胞 ,p53第 7,8外显子有点突变 ,而这些细胞都未出现典型的转化细胞形态特征的改变。结论 :反式 -BPDE可诱发人胎气管上皮细胞 p53基因点突变 ,此抑癌基因的点突变早于细胞形态学的改变。
Objective: To investigate the role of benzo (a) pyrene in the development of human lung cancer. METHODS: Human fetal tracheal epithelial cells cultured in vitro were transplanted with trans-BPDE, a metabolite of benzo (a) pyrene for 8 to 12 weeks. PCR-SSCP was used to detect the expression of p53 tumor suppressor genes 6,7,8 Point mutations in exons. RESULTS: Transfected-BPDE-treated human fetal bronchial epithelial cells showed a slight mutation in the exons 7 and 8 of p53, and none of these cells showed typical morphological changes of transformed cells. CONCLUSION: Trans-BPDE can induce p53 gene mutation in human fetal tracheal epithelial cells. The point mutation of this tumor suppressor gene is earlier than that of cell morphology.
