为探讨人脐静脉内皮细胞条件培养基(EC-CM)对人肝癌细胞(MHCC97H)癌干细胞表型的影响,分别培养人脐静脉内皮细胞和人肝癌细胞,收集EC-CM与MHCC97H细胞共培养,然后进行药物敏感、克隆形成能力、迁移/侵袭能力、癌干细胞标志分子表达以及肿瘤球形成能力分析。实验结果发现,EC-CM明显提高了MHCC97H对抗癌药物五氟尿嘧啶(5-FU)和顺铂(Cis)的耐药性,增加了MHCC97H细胞的集落形成能力。Transwell实验证实EC-CM不仅增强了MHCC97H细胞的迁移能力,也明显促进其侵袭能力。流式细胞仪分析发现EC-CM能上调MHCC97H细胞CD133的表达。与EC-CM共培养后,MHCC97H细胞形成肿瘤球的能力明显增强。研究结果表明,EC-CM能促进MHCC97H细胞的癌干细胞表型。
To investigate the effect of human umbilical vein endothelial cell conditioned medium (EC-CM) on the phenotype of human hepatocellular carcinoma (MHCC97H) cancer stem cells, human umbilical vein endothelial cells and human hepatocellular carcinoma cells were cultured and co-cultured with EC-CM and MHCC97H cells , Followed by drug sensitivity, clonogenicity, migration / invasion ability, expression of cancer stem cell marker molecules, and tumor sporulation ability analysis. The results showed that EC-CM significantly increased the resistance of MHCC97H cells to 5-fluorouracil (5-FU) and cisplatin (Cis) and increased the colony-forming ability of MHCC97H cells. Transwell experiments confirmed that EC-CM not only enhanced MHCC97H cell migration, but also significantly promote its invasive ability. Flow cytometry analysis found that EC-CM can up-regulate CD133 expression in MHCC97H cells. After co-cultured with EC-CM, the ability of MHCC97H cells to form tumor spheres was significantly enhanced. The results show that EC-CM can promote cancer stem cell phenotype MHCC97H cells.
