目的研究MIF4GD在肝细胞肝癌(HCC)中的表达及其对HCC预后判断的价值。方法 Western blot检测8例HCC及癌旁新鲜组织中MIF4GD表达。免疫组织化学法检测108例HCC及癌旁切片组织中MIF4GD及Ki-67表达。分析MIF4GD表达与HCC各临床病理参数间的相关性,并对病例进行随访,log-rank单因素和多因素Cox比例风险回归模型分析MIF4GD与HCC预后的关系。结果 MIF4GD在HCC组织中的表达明显低于相应的癌旁组织。MIF4GD的表达强度与HCC组织学分级、静脉侵袭和肝硬化状态相关(P<0.05),且MIF4GD表达与Ki-67呈负相关(r=-0.341,P<0.01)。MIF4GD低表达与HCC患者的不良预后显著相关(P<0.05)。结论MIF4GD在HCC中低表达,其可作为HCC独立的预后指标,提示MIF4GD可能与HCC的发生、发展密切相关。
Objective To study the expression of MIF4GD in hepatocellular carcinoma (HCC) and its value in predicting the prognosis of HCC. Methods Western blot was used to detect the expression of MIF4GD in 8 cases of HCC and adjacent non-cancerous tissues. Immunohistochemistry was used to detect the expression of MIF4GD and Ki-67 in 108 cases of HCC and adjacent normal tissues. The correlation between MIF4GD expression and clinicopathological parameters of HCC was analyzed. The relationship between MIF4GD and the prognosis of HCC was analyzed with follow-up, log-rank univariate and multivariate Cox proportional hazards regression model. Results The expression of MIF4GD in HCC tissues was significantly lower than that in corresponding paracancerous tissues. The expression of MIF4GD was correlated with HCC histological grade, venous invasion and cirrhosis (P <0.05), and the expression of MIF4GD was negatively correlated with Ki-67 (r = -0.341, P <0.01). The low expression of MIF4GD was significantly associated with poor prognosis in patients with HCC (P <0.05). Conclusions MIF4GD is low expressed in HCC, which may serve as an independent prognostic indicator of HCC, suggesting that MIF4GD may be closely related to the occurrence and development of HCC.
