目的:观察活化B淋巴细胞与NS1骨髓瘤细胞融合瘤苗对荷NS1瘤BABL/C小鼠免疫治疗作用。方法:用灭活NS1瘤细胞及其裂解物免疫BALB/C小鼠活化的B淋巴细胞;以50%PEG为融合剂,将免疫脾B淋巴细胞与野生型NS1骨髓瘤细胞融合,经HAT选择性培养、筛选融合细胞,以融合细胞作为瘤苗,免疫治疗荷NS1瘤小鼠2次。结果:融合细胞体在宿主体内的致瘤性降低;融合细胞瘤苗免疫治疗荷瘤小鼠的肿瘤组织体积逐渐减小、最终完全消失,小鼠存活期显著延长。结论:活化的B淋巴细胞与NS1细胞融合瘤苗能有效诱导小鼠体内的抗肿瘤反应。
Objective: To observe the immunotherapy effects of activated B lymphocytes and NS1 myeloma cell fusion vaccine on BALB/c mice bearing NS1 tumors. METHODS: BALB/c mouse activated B lymphocytes were immunized with inactivated NS1 tumor cells and their lysates; fusion splenic B lymphocytes were fused with wild-type NS1 myeloma cells using 50% PEG as a fusion agent and selected by HAT. The cells were cultured, screened for fusion cells, and fused cells were used as tumor vaccines to immunologically treat mice bearing NS1 tumors twice. Results: The tumorigenicity of the fused cell body in the host was reduced. The tumor tissue volume of the tumor-bearing mice treated with the fused cell tumor vaccine gradually decreased and eventually completely disappeared, and the survival time of the mice was significantly prolonged. CONCLUSION: The fusion of activated B lymphocytes with NS1 cells can effectively induce antitumor responses in mice.
