目的:观察极低密度脂蛋白受体(VLDL-R)的两种亚型在糖尿病患者脂肪组织中的分布和表达,以探讨其变化及意义。方法:选择10例正常人及9例糖尿病患者,手术中采集其腹壁及肠脂垂脂肪组织,从脂肪组织提取的mRNA用半定量-PCR方法扩增。结果:①正常腹壁及肠脂垂脂肪组织VLDLⅠ,Ⅱ型受体均有表达,两部位各亚型受体比较,差异无显著性意义(腹壁t=-0.454,P=0.6499;肠脂垂t=0.870,P=0.3843)。②糖尿病患者腹壁及肠脂垂脂肪组织中,Ⅰ型受体的分布和表达量与正常相比无明显变化,差异无显著性意义(腹壁t=-0.082,P=0.9349;肠脂垂t=-0.327,P=0.7439)。但Ⅱ型受体表达明显减少甚至丢失,与正常相比差异有显著性意义(腹壁t=3.576,P=0.0003;肠脂垂t=2.597,P=0.0094)。结论:糖尿病代谢紊乱影响VLDL-R表达及亚型分布。
Objective: To observe the distribution and expression of two subtypes of very low density lipoprotein receptor (VLDL-R) in adipose tissue of diabetic patients, and to explore its change and significance. Methods: Ten normal subjects and nine patients with diabetes mellitus were selected. The abdomen wall and intestinal adipose tissue were collected during operation. The mRNA extracted from adipose tissue was amplified by semi-quantitative PCR. Results: ① The expressions of VLDL Ⅰ and Ⅱ receptors in normal abdominal wall and intestinal lipoperoxid adipose tissue were all expressed. There was no significant difference between the two subtypes of receptor (t = -0.454, P = 0.6499, = 0.870, P = 0.3843). ② There was no significant difference in the distribution and expression of type I receptor in the abdominal wall and intestinal adipose tissue between diabetic patients and normal adipose tissue (t = -0.082, P = 0.9349, t = -0.327, P = 0.7439). However, the expression of type Ⅱ receptor was significantly reduced or even lost, with significant difference compared with normal (abdominal wall t = 3.576, P = 0.0003; intestinal obesity t = 2.597, P = 0.0094). Conclusion: The metabolic disorders of diabetes affect VLDL-R expression and subtype distribution.
